Introduction
St. John’s Wort is the dried herb of Chinese St. John’s Wort (Hypericum performatum) of the Garcinia Family. It is a wonted applications medicine with more than 2000 years of history. It was collected Zhong Yao Da Ci Dian and The Chinese Material Medica and some of local journals with “cleaning away heat and toximaterial, astringing to arrest bleeding, removing dampness by diuresis” function for traditional treatment of “hemptysis, hemoptysis, hematochezia, trauma bleeding, osteodynia due to pathogenic wind-dampness, foul breath sore, pyogenic infections, scald burn”.
St. John’s Wort Extract has been widely used in many countries and areas such like European and America. In German it has been taken as antidepression agent for about one hundred years and now is an OTC drug for antidepression and aided treatment AIDS. In American it was taken as dietary supplement for a number of diseases especially for depression. St. John’s Wort is collected in many pharmacopoeia and will continue to expand in the medicine field with further researches.
Chemical composition
The spectrum of constituent varies significantly due to numerous factors such like species difference, harvesting and handing practices, the specific part of the plant that is collected. The pharmacological activities of St. John’s Wort extract are due to a variety of constituents. We have assayed the content of hypericin in different species and different parts. We draw a conclusion that hypericin in St. John’s Wort is higher than other species and mainly insists in the flowers and leaves, especially in the flowers, then following by the fruits, but low in the stem and little in the radix. Pharmacological Activity
1. Anti-depression Activity
St. John’s Wort Extract is as a anti-depression agent in European (especially in German). Napythodianthrones, hyperforin and hypericin have strong anti-depressive activity. Anti-depressive mechanism is relation to the following actions:
ⅰ)Blocking the resorption of serotonin
ⅱ)Slight blocking of monoamine oxidase(MAO)
ⅲ)Blocking catechol-O-methyltransferase (COMT)
ⅳ)Interactions with the dopaminergic system and GABA receptors
2. Antiviral activity
In vitro hypericin and pseudohypericin inhibit human immunodeficiency virus (HIV) and other antiretrovirus as well as sindbis virus, murine cytomegalovirus, parainfluenza virus and herps simplex virus type I and type II by direct antiviral effects.
3. Anticancer Activity
Hypericin and hyperin are photosensitive. When they are activated by photo, they inhibited microchondria succinoxidase so that caused cancer cells irreversible damaged by photo.
4. Anti-inflammatory Activity
Biflavonoids in St. John’s Wort Extract has anti-inflammatory activity by activating white blood cell such as mononuclearmacrophage.
5. Antibiotic Activity
Low concentration of hyperforin in St. John’s Wort Extract inhibits the growth of Staphylococcus aures, Bacillus diphtheriae and MRSA.
Biological activities
St. John’s Wort has been one of the most popular plants. Its mainly use is for depression with describing clinically as “depression syndrome”: apathy, indifferent, dejection., lack of social contact, disturbances of sleep, appetite, etc.
Recently II period clinical trial of anti-AIDS of hypericin and pseudohypericin has been accomplished. Clinical trial of treatment of maliganoma is in process. Hypericum was widely used in the folk medicines of China and a number of European countries as a soothing agent, and an anti-phlogistic for treatment of inflammation of the bronchi, and the urogenital tract, a haemorrhoid treatment, a healing agent in the treatment of traumas, burns and scalds, ulcers of various kinds and other local general illnesses.
St. John’s Wort Extract has phototoxic activity that caused hemolysis and epileptic attack. So epilepsy patients are cautious to use.
